چکیده

The regulation of the mitotic histone HI kinase activity has been analyzed during the naturally synchronous cell cycle of Physanun polycephalum plasmodia. The universal binding property of the pl3 Schizosaccharomyces pombe gene product was used to precipitate and assay the cdc2 histone HI kinase activity. The kinase activity peaks at the beginning of metaphase and its decline, which requires protein synthesis, appears to be an early event during the metaphase process. Microtubular poisons, temperature shifts and DNA synthesis inhibitors were used to perturb cell cycle regulatory pathways and characterize their effects on cdc2 kinase activation. Our results suggest that the full activation of the mitotic kinase requires at least two successive triggering signals involving microtubular components and DNA synthesis.

ADP-ribosyltransferase in isolated nuclei during the cell cycle of Physarum polycephalum.

ADP-ribosyltransferase was measured in isolated nuclei of Physarum polycephalum. Activity was determined with and without exogenous DNA and histones. During the synchronous cell cycle the activity measured with exogenous substrates exhibited a typical peak enzyme pattern with a maximum of activity in S-phase, whereas activity measured without exogenous substrates displayed a step enzyme pattern...

Synchrony in the rhythm of the contraction-relaxation cycle in two plasmodial strands of Physarum polycephalum.

Rhythmicity of contraction of plasmodial strands of Physarum polycephalum was studied, by measuring the isometric tension exerted by isolated segments of the strands. When 2 strands were connected by way of a plasmodial mass, the contraction-relaxation cycle of the 2 strands synchronized. Such cycle activity of the strand was found to be well coordinated with shuttle streaming in the plasmodial...

Amphibian oocyte maturation induced by extracts of Physarum polycephalum in mitosis

The orderly progression of eukaryotic cells from interphase to mitosis requires the close coordination of various nuclear and cytoplasmic events. Studies from our laboratory and others on animal cells indicate that two activities, one present mainly in mitotic cells and the other exclusively in G1-phase cells, play a pivotal role in the regulation of initiation and completion of mitosis, respec...

Pteridine biosynthesis and nitric oxide synthase in Physarum polycephalum.

Physarum polycephalum, an acellular slime mould, serves as a model system to study cell-cycle-dependent events since nuclear division is naturally synchronous. This organism was shown to release isoxanthopterin which is structurally related to tetrahydrobiopterin, a cofactor of aromatic amino acid hydroxylases and of nitric oxide synthases (NOSs) (EC 1.14.13.39). Here, we studied Physarum pteri...

Nuclear acidic protein changes during differentiation in Physarum polycephalum.

A class of acidic nuclear phosphoproteins has been isolated throughout the mitotic cycle and at two points during differentiation in the slime mold Physarum polycephalum. The electrophoretic profiles of these proteins are reproducible and unchanging throughout the mitotic cycle, but reproducible changes occur during differentiation. The proteins are rapidly synthesized after mitosis, and their ...
